Effect of Paclitaxel in the Water Dynamics of MCF-7 Breast Cancer Cells Revealed by Dielectric Spectroscopy

Using dielectric spectroscopy experiments performed at multiple temperatures and frequency ranges, we demonstrate how the chemotherapy drug paclitaxel changes the dynamic properties of water in a breast cancer cell line (MCF-7). From the measured data, we present evidence that treatment with paclitaxel leads to a slight increase in activation energy in a relaxation related to bulk-like water. More importantly, we also observe that paclitaxel changes the constraining imposed by the biological interfaces on hydration water, whose single-particle dynamics becomes slower and with higher activation energy. These variations are only observable after freezing the dynamics from other cellular components, such as proteins and DNAs, regardless of the state of the cells, that is, treated or not treated or even if the cells are no longer viable. Therefore, changes in water dynamics could be detected prior to those related to the global dynamics within the cellular environment. They acknowledge the financial support from the European Union’s Horizon 2020 research and innovation programme (grant agreement 731019—EUSMI). Background Infection control depends on adequate microbe recognition and cell activation, yet inflammatory response may lead to organ dysfunction in sepsis. the aims of this study were to evaluate cell activation in the context of sepsis and its correlation with organ dysfunction. Methods A total of 41 patients were prospectively enrolled: 14 with sepsis, 12 with severe sepsis and 15 with septic shock. A total of 17 healthy volunteers were included as a control group. Patients were admitted to the Intensive Care Units and Emergency Rooms of Hospital São Paulo ( Federal University of São Paulo) and Hospital Santa Marcelina, São Paulo, Brazil. Toll- like receptor ( TLR) 2, TLR4, CD11b, CD11c and CD66b expression on neutrophil surfaces and oxidative metabolism measured by non- fluorescent dichlorofluorescein ( DCFH) oxidation in neutrophils and monocytes, using whole blood, were evaluated using flow cytometry. Organ dysfunction was measured using the sepsis- associated organ failure assessment ( SOFA) score. Results TLR2 expression on neutrophils was found to be downregulated in septic shock patients compared to healthy volunteers ( p = 0.05). No differences were found in CD11b and CD11c expression. CD66b expression was increased in the patient group compared to the control group ( p = 0.01). Neutrophil and monocyte oxidative burst was increased in septic patients compared to the control group at baseline and after stimulation with phorbol myristate acetate ( PMA), formylmethionylleucyl- phenylalanine ( fMLP), lipopolysaccharide ( LPS) and Staphylococcus aureus ( p 7 was higher than in patients with SOFA scores < 7, both in neutrophils and monocytes. However, oxidative burst in patients with sepsis was as high as in septic shock. Conclusion Surface receptors expression on neutrophils may be modulated across the continuum of sepsis, and enhanced or decreased expression may be found depending on the receptor considered. ROS generation is upregulated both in neutrophils and monocytes in septic patients, and it is differently modulated depending on the stage of the disease and the stimuli used.

Accounting Choices: The Brazilian Case Of Real Estate Investment

The objective in this study is to show that the business model of publicly traded real estate management companies in the Brazilian market explains their accounting choices to measure the investment properties. Therefore, the five main Brazilian investment property managers were studied, based on a method that triangulated documentary research, semistructured interviews and consistency analysis. The results of the analysis of the secondary data from the documentary research of the two main Brazilian companies in the real estate exploitation sector (one of which chose the fair value and the other the cost), together with the results of the interviews with the companies’ main internal and external agents direct or indirectly involved in the measuring choices of investment properties, indicated that the companies’ distinguished business model explains their distinct accounting choices to measure the investment properties. To enhance the consistency of these findings, next, the business models of the three consecutive largest Brazilian investment property managers were analyzed (two of which chose the fair value and one the cost), confronting them with their accounting choices; consistency was observed between the business models and the accounting choices, in accordance with earlier results. Although generalization of the results is not possible due to the small number of companies studied and the specific sector, evidence indicates that an integrated set of factors, which are considered in isolation in the literature, can explain the accounting choices. Several mutually interacting factors that explain the accounting choices, to the detriment of isolated factors, can contribute to expand the knowledge on this theme

Correction to: Ecosystems https://doi.org/10.1007/s10021-022-00801-5 Correction Published: 10 March 2023Coastal wetlands are key in regulating coastal carbon and nitrogen dynamics and contribute significantly to climate change mitigation and anthropogenic nutrient reduction. We investigated organic carbon (OC) and total nitrogen (TN) stocks and burial rates at four adjacent vegetated coastal habitats across the seascape elevation gradient of Cádiz Bay (South Spain), including one species of salt marsh, two of seagrasses, and a macroalgae. OC and TN stocks in the upper 1 m sediment layer were higher at the subtidal seagrass Cymodocea nodosa (72.3 Mg OC ha−1, 8.6 Mg TN ha−1) followed by the upper intertidal salt marsh Sporobolus maritimus (66.5 Mg OC ha−1, 5.9 Mg TN ha−1), the subtidal rhizophytic macroalgae Caulerpa prolifera (62.2 Mg OC ha−1, 7.2 Mg TN ha−1), and the lower intertidal seagrass Zostera noltei (52.8 Mg OC ha−1, 5.2 Mg TN ha−1). The sedimentation rates increased from lower to higher elevation, from the intertidal salt marsh (0.24 g cm−2 y−1) to the subtidal macroalgae (0.12 g cm−2 y−1). The organic carbon burial rate was highest at the intertidal salt marsh (91 ± 31 g OC m−2 y−1), followed by the intertidal seagrass, (44 ± 15 g OC m−2 y−1), the subtidal seagrass (39 ± 6 g OC m−2 y−1), and the subtidal macroalgae (28 ± 4 g OC m−2 y−1). Total nitrogen burial rates were similar among the three lower vegetation types, ranging from 5 ± 2 to 3 ± 1 g TN m−2 y−1, and peaked at S. maritimus salt marsh with 7 ± 1 g TN m−2 y−1. The contribution of allochthonous sources to the sedimentary organic matter decreased with elevation, from 72% in C. prolifera to 33% at S. maritimus. Our results highlight the need of using habitat-specific OC and TN stocks and burial rates to improve our ability to predict OC and TN sequestration capacity of vegetated coastal habitats at the seascape level. We also demonstrated that the stocks and burial rates in C. prolifera habitats were within the range of well-accepted blue carbon ecosystems such as seagrass meadows and salt marshes.17 página
